Past experiments have shown an antidepressant-like effect in mice administered sigma-1 receptor agonists and attenuation of these effects with sigma receptor antagonists . These receptors are found throughout the nervous system, and are concentrated in the hippocampus, frontal cortex, and olfactory bulb, consistent with a possible role in depression . Harmine acts to decrease synaptic glutamate via increased GLT-1/EAAT2 expression and subsequently increasing glutamate transport .

Previously it has been suggested that neuroticism could lead to difficult experiences when taking a psychedelic (Barrett et al., 2017), though other studies did not show this relation (Studerus et al., 2012; Haijen et al., 2018). The fact that personality can change after a psychedelic experience has been shown previously (Griffiths et al., 2011; MacLean et al., 2011; Bouso et al., 2015; Johnstad 2021), however, most of the studies showed changes in openness which were not found in the present study. As hypothesized, the study found decreases in neuroticism 1 week after the ayahuasca ceremony; the other four traits were unaffected.

The ayahuasca fanatics might say that nothing wrong can come out of the plant and from its experience. But I had to learn to experience and view life through a different perspective. But the depression, anxiety and panic attacks were still on full power. In a matter of hours, I went from being fine and normal, to experiencing the worst depression that I had ever experienced, something that felt unreal.
